If you want to disable USB port then this Tutorial Helps you a lot...
There are many Trick to Disable USB Port one of the Best is using Software of it.
But i want to share with you in Different Method...

Process:
-reboot the System (restart computer)
-Press "DEL" or F2 Button (on Keyboard) to go BIOS.
-In BIOS set up go to Standard (intergrat)Peripheral.
-Check USB port
-Then Change System in to Disable
-Press "F10" For Save & Exit.

Lks very good. But would like to add one mre thing: the key combination may change depending on different BIOS & respective manual should be consulted in this matter.

Thanks for this. Well some software like USB guarding software do have features like disabling USB port.

First of all right click on My computer then click on properties then hardware and then device manager. In device manager you can see at bottom Universal serial Bus controllers from here you can change your usb port status disable or enable. This is the best method according to my experience .
