Yes i think this was the base of the first ever coded bots, however, people used to get certain clothes and that confused the bots into following the player, where they could lure them for their inventory.

It would be possible, but it will be very hard to make it good. Also as far as I know you can't make a second mouse in vb.Net to do the clicks, also the anti-ban, antirandoms etc will be almost impossible.

The mouse problem is easily handled. When players install the program, it would also install a copy of Virtual Machines. When they run the bot, it will run inside a VM, so that the mouse clicks don't mess with your personal gameplay.
As for the anti-randoms. You are right. They are constantly added and removed. But it is possible to handle.
